4. Conclusion SP1049C has a novel mechanism of action based on the altered transport of the drug and its intracellular distribution. The increased efficacy of this product compared to doxorubicin, particularly in drug resistant settings, is due to: i ; increase in the drug influx; ii ; inhibition of the energy-dependent efflux; and iii ; changes in intracellular trafficking, as discussed above. As a result, a SP1049C binds with DNA ten times more actively than doxorubicin. The ability of SP1049C to kill multidrug resistant cells more effectively than their drug sensitive counterparts distinguishes this product from other doxorubicinbased formulations. This specific property of the product suggests that SP1049C may have the potential to be used not only for treatment of primary and relapsed tumours, but also for prevention of multidrug resistance development. Hypersensitisation of drug-resistant cells has not been reported by any of the main existing doxorubicin-enhancing strategies, i.e. i ; use of resistance-modulating agents Vrp, cyclosporins ii ; liposomal delivery Doxil iii ; chemical linkage to polymers PK-1, PEG-polyaspartate-doxorubicin ; .
